Port-Louis se compose d'une citadelle et d'une ville fortifiée. C'est Louis XIII qui fit reconstruire l'ancienne citadelle de Blavet, détruite, et développa la ville rebaptisée Port-Louis. La citadelle fut achevée de 1616 à 1622. Aux pieds du rempart sont installées un certain nombre de salles souterraines (entrepôts de vin, etc.). En 1636, Richelieu fit construire une demi-lune à l'extrémité du grand port, achevée par Charles de la Meilleraye en 1643. L'enceinte urbaine fut bâtie entre 1649 et 1653. Peu d'évolutions se sont produites entre le 17e et le 19e siècle, époque où des transformations importantes ont été pratiquées sur une partie des remparts et bastions (avec entre autres, l'établissement en 1850 d'une rampe d'accès qui reliait la porte de la Pointe à la place aux canons). Située face à la tour de Nesmond, la grande poudrière a été bâtie en 1750-1752, sur les plans de l'ingénieur Félix-François Le Royer de la Sauvagère, par l'entrepreneur des fortifications royales Jacques Robinet, dit Bourbonnais et son associé Gallen. François Gourier du Rocher exécuta le motif décoratif placé sur le pignon d'entrée sud : les armes de la duchesse de Mazarin, qui furent grattées en 1792. Ce bâtiment rectangulaire en pierre de taille de granit, est contenu dans une enceinte, sa voûte de couvrement présente un berceau maçonné en briques. Il conservait toutes les poudres des forts environnants ainsi que celles des vaisseaux faisant escale à Port-Louis ; sa capacité de stockage était de 120 000 livres.
