Le Grand Séminaire est installé en 1805, à la faveur du Concordat, dans un ancien monastère du XVIIIe siècle. Des campagnes de travaux successives tout au long du XIXe siècle dotent le séminaire d’une nouvelle façade alignée sur le rue du Hamel au Nord. Fermé à la suite de la loi de séparation de l’Église et de l’État, le Grand Séminaire est partagé entre les locaux de la Caisse des Dépôts & Consignations et d’une école de rééducation professionnelle pour les mutilés de la guerre (ainsi que par un hôpital pendant le premier conflit mondial), et désormais entre le CROUS et l’EPNAK. L'histoire du bâtiment est marquée par ses différentes fonctions depuis sa construction, d'abord religieuse (ancien couvent des capucins), ensuite politique (lieu de rassemblements révolutionnaires) et économique (atelier de la monnaie, filature, traite négrière) et enfin hospitalière (hôpital de chirurgie orthopédique).

L’ancien Grand Séminaire présente une organisation en H héritée de la reconstruction du bâtiment au XVIIIe siècle, avec deux cours au Sud-Ouest et au Nord-Est, cette seconde cour étant entourée par une galerie portée par une colonnade. La construction d’une nouvelle façade principale du séminaire donnant sur la rue du Hamel (seconde moitié du XIXe siècle avant un agrandissement au début du XXe) a donné à cette cour une forme de flèche. La chapelle néo-gothique du séminaire, construite entre 1874 et 1875, possède des dimensions imposantes avec un important programme sculpté. Cette chapelle comporte en outre cinq chapelles absidiales encore ornées d’un décor peint.
