Lectoure came into the sphere of influence of the Counts of Armagnac in the mid-14th century, and they installed their castle-residence to the west of the spur which dominates the town, on the remains of a pre-existing castle. The castle suffered significant destruction during the siege of the town by Louis XI (1472-1473). Becoming royal property, it was repaired, then underwent works of improvement until the 17th century. Between 1583 and 1590, it was separated from the town by a double bastion. Part of the premises served as a prison. In 1758, the bishop Claude-François de Narbonne-Pelet asked Louis XV to cede the castle to him in order to establish a hospital-manufactory intended to replace the Holy Spirit hospital. Having obtained a favourable reply, he had a vast building of U-shaped plan constructed from 1758 to 1766. The bastion separating the castle from the town was removed. Only the western end of the castle was retained to house the pharmacy and the dependencies of the hospital. The north bastion of the castle collapsed shortly after 1824. The south wing and the chapel of the hospital were completed between 1808 and 1812; the chapel was decorated by the Lectoure painter Lasseran. New buildings were erected in the 19th century, to the north and south, and the hospital was finally decommissioned in 2013.
