L'existence de l'hôpital est attestée dès le 13e s. L'hôpital Saint-Laurent est créé en 1633 grâce à la générosité du procureur du Roi, Guillaume Labye. Mais la véritable fondation des hospices commence avec Antide Midan, prêtre de Nuits qui lui lègue tous ses biens. C'est à partir de 1692 que l'hôpital s'installe sur son emplacement actuel et occupe progressivement le terrain situé entre la rue Henri-Challand et le Meuzin. Plusieurs vagues de constructions répondent à l'évolution de la fréquentation de l'établissement et aux exigences de confort des malades : salle Saint-Laurent en 1692, salle Marie-Madeleine en 1739 (architecte Pierre Le Mousseux), nouvelle apothicairerie en 1813, salle Saint-Etienne et agrandissement de la chapelle en 1842 (Pierre-Paul Petit), salle Saint-Joseph en 1878 (Alfred Chevrot), pavillon Laënnec en 1934 (Emile Robert) pour les malades de la tuberculose. Dans la deuxième moitié du 20e siècle, des adjonctions modernes sont construites. Les biens de plusieurs léproseries ou maladreries de Nuits et des villages voisins réunis aux hospices en 1693, puis de nombreux dons jusqu'au milieu du 18e siècle lui assurent des ressources intéressantes. Les hospices conservent aujourd'hui un important domaine de 11 ha de vigne qui lui permet d'assurer l'entretien et la restauration des bâtiments.
